Quantitative real-time PCR for the clinical detection of Helicobacter pylori Genet. Mol. Biol.
Ribeiro,Marcelo Lima; Ecclissato,Christina Cunha; Mattos,Ricardo Gabriel; Mendonca,Sergio; Pedrazzoli Jr.,José.
Accurate diagnosis of Helicobacter pylori infection is very important in both clinical practice and research. We evaluated the sensitivity of real-time PCR (RT-PCR) for the detection and quantification of Helicobacter pylori using DNA from 91 human gastric biopsy samples divided into three groups: 46 biopsies from untreated patients who according to the references methods were considered H. pylori-negative (group A); 35 biopsies from patients previously treated against H. pylori and considered to be cured by "gold standard" tests (group B); and 10 biopsies from patients H. pylori-positive by all available methods (group C). The sensitivity of the RT-PCR assay was higher than that of standard methods. Of the 81 patients considered to be uninfected according...

Phylogenetic analysis, based on EPIYA repeats in the cagA gene of Indian Helicobacter pylori, and the implications of sequence variation in tyrosine phosphorylation motifs on determining the clinical outcome Genet. Mol. Biol.
Tiwari,Santosh K.; Sharma,Vishwas; Sharma,Varun Kumar; Gopi,Manoj; Saikant,R; Nandan,Amrita; Bardia,Avinash; Gunisetty,Sivaram; Katikala,Prasanth; Habeeb,Md. Aejaz; Khan,Aleem A.; Habibullah,C.M..
The population of India harbors one of the world's most highly diverse gene pools, owing to the influx of successive waves of immigrants over regular periods in time. Several phylogenetic studies involving mitochondrial DNA and Y chromosomal variation have demonstrated Europeans to have been the first settlers in India. Nevertheless, certain controversy exists, due to the support given to the thesis that colonization was by the Austro-Asiatic group, prior to the Europeans. Thus, the aim was to investigate pre-historic colonization of India by anatomically modern humans, using conserved stretches of five amino acid (EPIYA) sequences in the cagA gene of Helicobacter pylori. Simultaneously, the existence of a pathogenic relationship of tyrosine...

Relationship between Helicobacter pylori detection and an increased risk of infection in childhood J. Venom. Anim. Toxins incl. Trop. Dis.
Venerando,R; Rasmussen,LT; de Labio,RW; Gatti,LL; Francisco,O; Viani,GA; Rivera,LML; Payão,SLM.
The risk of developing gastric cancer is believed to be related to differences among Helicobacter pylori strains and the inflammatory responses mediated by host genetic factors. H. pylori infection is acquired at an early age and in the absence of appropriate antibiotic therapy, it generally persists for life. Tp53 gene regulates the transcription of several cytokines and chemokines involved in innate immunity and its action may be influenced by the presence of different H. pylori strains. The present study aimed to detect H. pylori in pediatric patients, to access Tp53 polymorphism at codon 72 and to correlate such findings with age and histopathological results. Three hundred and forty-two patients were analyzed. DNA from their gastric biopsies was...

tlpA gene expression is required for arginine and bicarbonate chemotaxis in Helicobacter pylori Biol. Res.
Cerda,Oscar A; Núñez-Villena,Felipe; Soto,Sarita E; Ugalde,José Manuel; López-Solís,Remigio; Toledo,Héctor.
About half of the human population is infected with Helicobacter pylori, a bacterium causing gastritis, peptic ulcer and progression to gastric cancer. Chemotaxis and flagellar motility are required for colonization and persistence of H. pylori in the gastric mucus layer. It is not completely clear which chemical gradients are used by H. pylori to maintain its position. TlpA, a chemotaxis receptor for arginine/ bicarbonate, has been identified. This study aimed to find out whether tlpA gene expression is required for the chemotactic response to arginine/bicarbonate. Wild-type motile H. pylori ATCC 700392 and H. pylori ATCC 43504, a strain having an interrupted tlpA gene, were used. Also, a tlpA-knockout mutant of H. pylori 700392 (H. pylori 700-tlpA::cat)...

Correlation between Helicobacter pylori infection, gastric diseases and life habits among patients treated at a university hospital in Southeast Brazil BJID
Módena,José Luiz Proença; Acrani,Gustavo Olszanski; Micas,André Fernando Ditondo; Castro,Marly de; Silveira,Wanderley Dias da; Módena,José Luiz Pimenta; Oliveira,Ricardo Brandt de; Brocchi,Marcelo.
Helicobacter pylori is considered a significant agent in the development of various gastric diseases. However, the diseases caused by this bacterium are known as being multi-factorial, with the genotype, immune system and life habits of the host playing important roles in the establishment of the clinical outcome. Also, H. pylori exhibit a high degree of genetic variability, contributing to the complexity of the host-pathogen relationship. These observations, considered together with the widely-varying origins and social habits of the Brazilian population, lead us to speculate about the influence of these life habits on H. pylori infection and the clinical outcome. Therefore, in this study we evaluated the relationship between H. pylori infection and...
